Celiac disease is an autoimmune disorder in which the immune system mistakenly attacks the lining of the small intestine in response to gluten — a protein found in wheat, rye, barley, and oats. Over time, this immune reaction damages the intestinal lining and can lead to serious nutritional deficiencies and health complications. Importantly, a gluten-free diet is not required before testing, as this test analyzes your DNA rather than antibody levels.
Almost all people with celiac disease carry at least one genetic risk allele in their HLA genes. This test detects nucleotide changes in three celiac-associated alleles of the HLA-DQA1 and HLA-DQB1 genes (HLA-DQA1*05, HLA-DQB1*02, HLA-DQB1*0302). These variants can pair to form proteins known as DQ2 and DQ8, which indicate varying levels of celiac disease risk.

Do I need to be eating gluten before taking this test?
No. Unlike antibody blood tests for celiac disease, this DNA test analyzes your genetic makeup rather than your immune response. Your results will be accurate regardless of whether you are currently following a gluten-free diet.
What does a negative result mean?
A negative result means no HLA celiac-associated alleles were detected. This rules out celiac disease with a very high degree of certainty, as fewer than 1 in 2,518 people without these alleles develop the condition.
Does a positive result mean I have celiac disease?
Not necessarily. Carrying HLA risk alleles increases your genetic susceptibility but does not confirm a diagnosis. Many people carry these variants without ever developing celiac disease. A positive result should be discussed with your physician, who may recommend further testing.
How is the DNA sample collected?
DNA is collected using a painless buccal (cheek) swab. Simply rub the swab gently inside your mouth against your cheek for 15 seconds. No needles, blood draws, or clinic visits are required.
